Cellular Assys in LOV Format
Shortly after its introduction the LOV technique (Ruzicka 2000) was integrated into instrumentation for cellular assays. While initially the LOV served only as a “front end” to fluorescence microscopy, as discussed on previous slides, it was soon realized that the use of optical fibers for probing a cellular culture directly within the LOV module has the following advantages:
- small volume of interrogated cells
- enclosed environment, protected from ambient air
- possibility of multi-parameter measurement
- protection from ambient light
Use of the LOV facilitated not only fluorescence-based monitoring, but opened the door to UV-VIS absorbance and light scattering measurements, as well as to voltammetry, and its combination with fluorescence for multi-parameter based assays.
